Preparation: Alcohol spray can with 75% medical disinfection alcohol, PBS buffer, serum-containing medium, pasteurized pipette, pipette, waste liquid tank.
Specific steps:
1. Put the equipment into the ultra-clean table and turn on the ultraviolet irradiation to sterilize. If the cultured cells are sensitive to temperature, you can put the serum-containing culture medium bottle in the 37℃ water bath to make the culture medium temperature 37℃, and then transfer Go to the ultra-clean bench for UV sterilization. In addition, the cell culture room also needs UV irradiation for about half an hour.
2. Take out the cell culture flasks from the incubator, spray 75% alcohol on the surface of the bottle with an alcohol spray can for disinfection, transfer to the ultra-clean table, open the lid, gently suck out the old culture medium, use a pipette to add fresh serum-containing medium, Be careful not to add the culture medium from the adherent surface of the cells, but from the side wall, gently, so as not to wash off the cells.
3. After adding the culture medium, close the lid and mark the bottle to indicate the time for changing the medium, cell type, operator and other information.
4. Before putting it into the incubator, spray the surface of the bottle with alcohol again, and then remember to tidy up the operating table, wipe the surface of the ultra-clean table with alcohol, and clean up waste liquid and garbage.
When changing the liquid, pay attention to sterility throughout the process. Operators should wear sterile clothes, sterile gloves and masks, and be careful when touching the mouth of the cell culture flasks with their hands to avoid contamination.
